چکیده

Lotus corniculatus, Leguminosae, O-Methyltransferase, Flavonols, 8-O-Methylation, 3'-0-Methylation Two O-methyltransferases specific for flavonol rings A and B were isolated from young flower buds of Lotus corniculatus. They were partially purified by ammonium sulphate precipitation and successive chromatography on Sephadex G-100 and Polybuffer ion exchanger. One enzyme focused at pi 5.5 and catalysed the O-methylation of position 8 of flavonols with a pH optimum of 8.1. The other enzyme had a pi of 5.1 and preferentially attacked position 3' at an optimum pH of 7.7. The methylated products of both enzymes seem to contribute to the flower colour of Lotus and may be used as biochemical markers in genetic studies of this genus.

The role and importance of DNA methylation in spermatogenesis process

Background: DNA methylation is one of the epigenetic marks that are created by de novo DNA methylation and be maintained through cell division. This process is catalyzed by DNA methyltransferases. DNA methylation establishment in germ line is important, since they have the potential to regulate gene expression in offspring and improper DNA methylation patterns in germ lines has serious conseque...

Functional Characterisation of Three O-methyltransferases Involved in the Biosynthesis of Phenolglycolipids in Mycobacterium tuberculosis

Phenolic glycolipids are produced by a very limited number of slow-growing mycobacterial species, most of which are pathogen for humans. In Mycobacterium tuberculosis, the etiologic agent of tuberculosis, these molecules play a role in the pathogenicity by modulating the host immune response during infection. The major variant of phenolic glycolipids produced by M. tuberculosis, named PGL-tb, c...

Engineering monolignol 4-O-methyltransferases to modulate lignin biosynthesis.

Lignin is a complex polymer derived from the oxidative coupling of three classical monolignols. Lignin precursors are methylated exclusively at the meta-positions (i.e. 3/5-OH) of their phenyl rings by native O-methyltransferases, and are precluded from substitution of the para-hydroxyl (4-OH) position. Ostensibly, the para-hydroxyls of phenolics are critically important for oxidative coupling ...

A Conformational Switch in the Active Site of BT_2972, a Methyltransferase from an Antibiotic Resistant Pathogen B. thetaiotaomicron

Methylation is one of the most common biochemical reactions involved in cellular and metabolic functions and is catalysed by the action of methyltransferases. Bacteroides thetaiotaomicron is an antibiotic-resistant bacterium that confers resistance through methylation, and as yet, there is no report on the structure of methyltransferases from this bacterium. Here, we report the crystal structur...

Tobacco O-methyltransferases involved in phenylpropanoid metabolism. The different caffeoyl-coenzyme A/5-hydroxyferuloyl-coenzyme A 3/5-O-methyltransferase and caffeic acid/5-hydroxyferulic acid 3/5-O-methyltransferase classes have distinct substrate specificities and expression patterns.

The biosynthesis of lignin monomers involves two methylation steps catalyzed by orthodiphenol-O-methyltransferases: caffeic acid/5-hydroxyferulic acid 3/5-O-methyltransferases (COMTs) and caffeoyl-coenzyme A (CoA)/5-hydroxyferuloyl-CoA 3/5-O-methyltransferases (CCoAOMTs). Two COMT classes (I and II) were already known to occur in tobacco (Nicotiana tabacum) and three distinct CCoAOMT classes ha...
